After graduating, She appeared on the hit TV show, Cemberimde Gul Oya portraying the character of Zarife. This was her first television series to develop an acting career. Following, she was awarded 'Best Actress' in the Republic of Serbia and Montenegro International TV Festival as she played the best role of Gülizar, in the TV movie Gülizar.
Later in 2005, Buyukustun debuted her role in the television series Ihlamurlar Altinda, in the series she dedicated her life for to character, Filiz. This series became a huge hit all over the world.
Her successful television series are, Gonülcelen, 20 Minutes, Black Money Love, and currently, The Brave and the Beautiful. Besides, she also starred in numerous movies such as Sinav (Omer Faruk Sorak), My Father and My Son, (Cagan Irmak), Ask Your Heart (Yusuf Kurcenli), Memories of the Wind (Ozcan Alper), Tight Dress (Hiner Saleem).
For debuting as Melek, in 2013, Tuba was nominated for Best Actress at the "42 International Emmy Awards," and she was the first actress from Turkey to become the nominee of Emmy award.
